So I want to start to develop one game to give me a spoon look so the speak. But my question is what program should i Use to make my characters the course I was in just used the stupid fish sprites and were there already.

You can use the fish again or any number of placeholder characters from the web. It depends on the sort of game you'd like to develop.

Use whichever 2D art package you're most comfortable with. I use Paint Shop Pro and Photoshop, but some use Gimp and others use other apps. They all pretty much output the same file formats you'd use (JPG, PNG- my fav). Hell, you can even sketch the art and scan it in to use if you want to use that style. Hope that helps.
